چکیده
Background: The immunomodulatory effects of mesenchymal stem cells (MSCs) and their exosomes have been receiving increasing attention. This study investigated the immunoregulatory human amniotic (hAMSCs) on phytohemagglutinin (PHA)-induced peripheral blood mononuclear (PBMCs). Methods: hAMSCs used in experiment were identified by light microscopy flow cytometry, differentiation ability was determined Oil Red O Alizarin staining. expressions transforming growth factor (TGF)-β, indoleamine 2,3-dioxygenase (IDO), cyclooxygenase-2 (COX-2), hepatocyte (HGF), interleukin (IL)-6 detected quantitative real-time polymerase chain reaction western blotting. PBMCs, hAMSCs, collected for in vitro group culture. Then analyzed cytometry Enzyme-linked immunosorbent assay. Results: successfully extracted from membrane. TGF-β, IDO, COX-2, HGF, IL-6 significantly expressed hAMSCs. In co-culture showed that promoted proliferation Th2 PHA-induced while inhibited secretion TNF-α IL-4 IL-10, had more significant than exosomes. Conclusions: or could exert PBMCs affecting cell cytokine secretion.

Gene Expression Profile of Adherent Cells Derived From Human Peripheral Blood: Evidence of Mesenchymal Stem Cells
Mesenchymal stem cells (MSCs) provide a novel option in cellular therapy and tissue engineering. Recent studies indicated that it is possible to obtain MSCs from peripheral blood by attachment ability to plastic surface. To evaluate adherent cells derived from peripheral blood, their expression profile and surface markers were investigated. The results of RT-PCR indicated that these cells expre...
متن کاملOptimization of human peripheral blood mononuclear cells (PBMCs) cryopreservation
Cryopreservation is the method of choice for long term storage of human PBMCs. This study was designed to compare the different combinations of variables affecting the cryopreservation of PBMCs samples. The viability of PBMCs separated from 2×5 ml peripheral blood samples obtained from 16 healthy adult volunteers, were measured using trypan blue dye exclusion method just before freezing with ...
متن کاملComparison of Characteristics of Human Amniotic Membrane and Human Adipose Tissue Derived Mesenchymal Stem Cells
BACKGROUND Mesenchymal stem cells (MSCs) are ideal candidates for treatment of diseases. Amniotic membranes are an inexpensive source of MSCs (AM-MSC) without any donor site morbidity in cell therapy. Adipose tissue derived stem cells (ASCs) are also suitable cells for cell therapy. There is discrepancy in CD271 expression among MSCs from different sources. In this study, the characteristics...
متن کاملHuman Mesenchymal Stem Cells and Their, Clinical Aapplication
There are two main categories for stem cells a cording to their origin: Embryonic Stem Cells and Adult Stem Cell. Mesenchymal stem cell, supporting hematopoetic stem cells in bone marrow, can regenerate tissues such as bone, cartilage, muscle, tendon and fatty tissue. These cells were recognized for the first time by Friedenstein and Petrokova who could isolate theme from rat bone marrow.Mesenc...
متن کاملImmunoregulatory impact of human mesenchymal-conditioned media and mesenchymal derived exosomes on monocytes
Mesenchymal stem cells (MSCs) are well known due to their immunomodulatory effect, but the exact mechanisms have not been defined. Several studies demonstrated that the exerted immunoregulatory effect of these cells could be mediated by paracrine factors to illustrate, cytokines, chemokine, and among which, extracellular vesicles are one of them to play a crucial role. Moreover, it is assumed t...
